Application of microsatellite markers for breeding and genetic conservation of herds of Pantaneiro sheep Electron. J. Biotechnol.
Crispim,Bruno do Amaral; Seno,Leonardo de Oliveira; Egito,Andréa Alves do; Vargas Junior,Fernando Miranda de; Grisolia,Alexéia Barufatti.
Background The aim of the present study was to assess the genetic diversity of Pantaneiro sheep, using microsatellite markers, in order to assist maintenance and management plans, enhance mating systems and reduce the inbreeding rate. A total of 127 animals were genotyped at eight microsatellite loci. They belonged to populations from the Experimental Farm of the Universidade Federal da Grande Dourados (UFGD) (Dourados/MS/Brazil) and Embrapa Pantanal (Corumbá/MS/Brazil). Results The population of Pantaneiro sheep from the UFGD exhibited a high mean number of alleles (11.13) and allelic richness (10.66). The polymorphic information content was highly informative in the locus studied, resulting in a mean value of 0.71. Observed heterozygosity was lower than...

Detection and evaluation of selection signatures in sheep PAB
Paim,Tiago do Prado; Ianella,Patrícia; Paiva,Samuel Rezende; Caetano,Alexandre Rodrigues; Pimentel,Concepta Margaret McManus.
Abstract: The recent development of genome-wide single nucleotide polymorphism (SNP) arrays made it possible to carry out several studies with different species. The selection process can increase or reduce allelic (or genic) frequencies at specific loci in the genome, besides dragging neighboring alleles in the chromosome. This way, genomic regions with increased frequencies of specific alleles are formed, caracterizing selection signatures or selective sweeps. The detection of these signatures is important to characterize genetic resources, as well as to identify genes or regions involved in the control and expression of important production and economic traits. Sheep are an important species for theses studies as they are dispersed worldwide and have...

D-loop haplotype diversity in Brazilian horse breeds Genet. Mol. Biol.
Ianella,Patrícia; Albuquerque,Maria do Socorro Maués; Paiva,Samuel Rezende; Egito,Andréa Alves do; Almeida,Leonardo Daniel; Sereno,Fabiana T. P. S.; Carvalho,Luiz Felipe Ramos; Mariante,Arthur da Silva; McManus,Concepta Margaret.
Abstract The first horses were brought to Brazil by the colonizers after 1534. Over the centuries, these animals evolved and adapted to local environmental conditions usually unsuitable for exotic breeds, thereby originating locally adapted Brazilian breeds. The present work represents the first description of maternal genetic diversity in these horse breeds based on D-loop sequences. A D-Loop HSV-I fragment of 252 bp, from 141 horses belonging to ten Brazilian breeds / genetic groups (locally adapted and specialized breeds) were analysed. Thirty-five different haplotypes belonging to 18 haplogroups were identified with 33 polymorphic sites. Haplotype diversity (varying from 0.20 to 0.96) and nucleotide diversity (varying from 0.0039 to 0.0239) was lower...

Genetic diversity of captive spotted paca (Agouti paca) from south east Brazil assessed by the RAPD-PCR technique R. Bras. Zootec.
Antunes,Karine Vieira; Machado,Théa Mírian Medeiros; Serão,Nicola Vergara Lopes; Guimarães,Simone Eliza Facione; Paiva,Samuel Rezende.
The genetic diversity was analyzed among spotted paca (Agouti paca) from three commercial flocks located in Brazil. As the genome of this species is unknown, the RAPD-PCR technique was used. Ten primers generated sixty polymorphic bands. The among and within population genetic variability estimated by analysis of molecular variance (AMOVA) was 12.55 and 87.45%, respectively. The shortest Nei distance value was 11.76% among the Carangola (CG) and São Francisco do Glória (SF) populations. This value can be explained by the exchange of reproduction males and females between the two geographically close breeding sites. The analysis of principal components showed well defined and structured groups aggregating animals according their population of origin, with...

Genetic monitoring of a Santa Ines herd using microsatellite markers near or linked to the sheep MHC R. Bras. Zootec.
Petroli,César Daniel; Paiva,Samuel Rezende; Corrêa,Marilma Pachêco Chediak; McManus,Concepta.
This study aimed to analyze genetic diversity in a conservation nucleus of Santa Inês sheep using thirteen microsatellite loci on chromosome 20 (where the Sheep Major Histocompatibility Complex - Ovar-MHC - is found). Seventy three animals from one herd born from 2004 to 2006 were evaluated as a principal nucleus. Seventy one animals from two other herds were used as control comparison. There was a reduction in heterozygosity over the years in relation to the whole population. This may be due to the repeated use of the same sires. The estimates of molecular coancestrality also indicated an increase in genetic similarity between individuals with the herd over the years. A high number of alleles occurred exclusively in the principal nucleus herd, but with a...

Managing genetic diversity and society needs R. Bras. Zootec.
Mariante,Arthur da Silva; Egito,Andréa Alves; Albuquerque,Maria do Socorro Maués; Paiva,Samuel Rezende; Ramos,Alexandre Floriani.
Most livestock are not indigenous to Brazil. Several animal species were considered domesticated in the pre-colonial period, since the indigenous people manage them as would be typical of European livestock production. For over 500 years there have been periodic introductions resulting in the wide range of genetic diversity that for centuries supported domestic animal production in the country. Even though these naturalized breeds have acquired adaptive traits after centuries of natural selection, they have been gradually replaced by exotic breeds, to such an extent, that today they are in danger of extinction To avoid further loss of this important genetic material, in 1983 Embrapa Genetic Resources and Biotechnology decided to include conservation of...

Morphological measurements and body indices for Cuban Creole goats and their crossbreds R. Bras. Zootec.
Chacón,Edilberto; Macedo,Fernando; Velázquez,Francisco; Paiva,Samuel Rezende; Pineda,Eliecer; McManus,Concepta.
In this work, linear body measures were taken and production indices calculated for 100 Cuban Creole goats and 100 crossbred goats in order to aid in the characterization of animal genetic resources in Cuba. Low variation was found for all indices of the Creole goats, showing homogeneity between the groups of animals studied. Most of the functional indices are related to the milk biotype which is in agreement the possible origin of the breed from animals of the Iberian Peninsula and Canary Islands. The crossbreds were more varied, mostly due to undesigned disorganized crossing. These data may help in identifying a commercial niche for the breed and contribute to in situ conservation of the Cuban Creole goat.

Of Experts, Politicians and Beasts: Setting Priorities in Farm Animal Conservation Choices AgEcon
Fadlaoui, Aziz; Roosen, Jutta; Baret, Philippe V..
We consider the choice of farm animal breeds for conservation programmes. Based on an analysis of past decisions in EU member countries to enter breeds into the conservation programmes of rural development plans and based on the results of an expert survey among breed societies and scientists, we find an inconsistency in the valuation of breed characteristics. Policy makers seem to be less concerned about considering true extinction risk and diversity and more about cultural values and about means to benefit a larger number of farmers for raising rare breeds.

Validation of a customized subset of SNPs for sheep breed assignment in Brazil PAB
Paim,Tiago do Prado; McManus,Concepta; Vieira,Fábio Danilo; Oliveira,Stanley Robson de Medeiros; Facó,Olivardo; Azevedo,Hymerson Costa; Araújo,Adriana Mello de; Moraes,José Carlos Ferrugem; Yamagishi,Michel Eduardo Beleza; Carneiro,Paulo Luiz Souza; Caetano,Alexandre Rodrigues; Paiva,Samuel Rezende.
Abstract: The objective of this work was to evaluate the usefulness of a subset of 18 single nucleotide polymorphisms (SNPs) for breed identification of Brazilian Crioula, Morada Nova (MN), and Santa Inês (SI) sheep. Data of 588 animals were analyzed with the Structure software. Assignments higher than 90% confidence were observed in 82% of the studied samples. Most of the low-value assignments were observed in MN and SI breeds. Therefore, although there is a high reliability in this subset of 18 SNPs, it is not enough for an unequivocal assignment of the studied breeds, mainly of hair breeds. A more precise panel still needs to be developed for the widespread use in breed assignment.

Validation of a microsatellite panel for parentage testing of locally adapted and commercial goats in Brazil Genet. Mol. Biol.
Silva,Elizabete Cristina da; McManus,Concepta Margaret; Guimarães,Maria Pia Souza Lima Mattos de Paiva; Gouveia,Aurora M.G.; Facó,Olivardo; Pimentel,Daniel M.; Caetano,Alexandre Rodrigues; Paiva,Samuel Rezende.
Brazilian goats are generally kept in small herds and extensive rearing systems, mainly in the northeastern region of the country. Despite production improvement in recent years, the lack of pedigree control has affected genetic progress. This study aimed to validate a panel of 16 microsatellites for parentage testing in locally adapted and commercial goats breeds raised in Brazil, as well as to compare its efficiency with the panel recommended by the Brazilian Ministry of Agriculture, Livestock and Supplies (MAPA) in 2004. The number of alleles and expected heterozygosity (He) per marker ranged from four to 18, and from 0.051 to 0.831, respectively. Using all markers, 100% of parentage cases of the validation dataset were resolved with a strict confidence...
